soil

Sara Scheiding

I have been working in Dr. Kirsten Hofmockel's lab as part of the 2011 Biogeosciences Research Experiences for Undergrads program through Iowa State University. I have had an amazing summer learning so many new things about soil and getting to know some wonderful people. I recently moved to Ames, Iowa from Ellsworth Community College in Iowa Falls, Iowa following graduating with an Associate of Science in Pre-Engineering. I will continue my education at Iowa State University this fall pursuing a Bachelors degree in Industrial Engineering along with a Master of Business Administration.

Effect of nitrate and glucose addition on denitrification and nitric oxide reductase (cnorB) gene abundance and mRNA levels in Pseudomonas mandelii inoculated into anoxic soil

Abstract

Elevated temperatures and carbon dioxide concentrations- effects on selected microbial activities

Here is the Abstract from this paper:
Human activities have increased greenhouse gas concentrations in the atmosphere. Research has demonstrated this increased concentration will affect our climate

Relationships

mRNA extraction (CTAB, phenol-chloroform), DNA extraction

Additional papers with mRNA extraction techniques, including
* Hurt&Tiedje.etal_2001: CTAB extraction, simultaneous extraction of DNA & RNA for transcription analysis
* McGrath.etal_2008: Specifically processed for mRNA using MoBio (phenol-chloroform) extraction; pre-processing in field
* Poretsky.etal_2005: mRNA extraction environmental samples

DNA extraction comparison: Evaluates different methods for DNA extraction in different soils

Qiagen extraction reagents

The following papers both used Qiagen reagents to extract mRNA from soil samples. One was unsuccessful in amplifying, perhaps because soils were stored at -20.
